Piccolo can be obtained at any time after completing Chapter 5.  Visiting Merlee in Flopside will prompt a fetch quest to see Merluvlee, Bestovius, Watchitt, and Merlumina in that order, and then again in reverse.  Once Merlee has received the item she requested, she gives Mario and co. a key to the locked house in Flopside where Piccolo is found.

Piccolo's card can be found on the first floor of the Flopside Outskirts.  Near the seventh Heart Pillar is a wall that only Luigi can jump onto. While on this wall, Mario can switch to 3-D to find a path to the other side of the room where this chest is found.  It can be sold for 120 coins.
